According to http://windows.microsoft.com/en-US/windows-vista/What-is-safe-mode, Safe Mode is a "troubleshooting option for Windows that starts your computer in a limited state."  Before attempting to access Safe Mode with Window-Eyes, you will want to make certain that you are using at least Window-Eyes 8.0 Beta 1 or higher and that you have enabled audio support in Safe Mode.  To enable audio support for Window-Eyes in Safe Mode, please perform the following steps:

1.    Press Control-Backslash to open Window-Eyes
2.    Press Alt-A to open the Apps menu
3.    Press the Up or Down Arrow to navigate to Windows Enhanced
4.    Press Enter
5.    Down Arrow to Enable Audio Support in Safe Mode
6.    Press Enter

Note: If you are using Vista or above, you will need to accept the User Account Control (UAC) if UAC is turned on.

With this change made, you can now start Windows in Safe Mode and access this troubleshooting environment with Window-Eyes.  It should be noted that you may need sighted assistance to start your computer in Safe Mode.
